DNA-protein crosslinks (DPCs) are common DNA lesions caused by widely used chemotherapeutics, like cisplatin, but also by byproducts of the cellular metabolism, such as aldehydes. If left urepaired DPCs are likely to promote genome instability, predisposing for cancer and aging. Using Xenopus egg extract as a model system, I aim to identify the key factors required for processing DPCs, and to delineate their mechanisms of action. This will be instrumental for understanding how a very common and toxic lesion like a DPC is recognized and repaired also in humans.
