N2 - The main objective of this thesis is to develop quantitative fluorescence-based,spectroscopic tools for probing the 3D structure and dynamics of DNA andRNA. The thesis is founded on six peer-reviewed papers covering mainly thedevelopment, characterization and use of fluorescent nucleobase analogues. Inaddition, four software packages is presented for the simulation and quantitativeanalysis of time-resolved and steady-state UV-Vis absorption and fluorescenceexperiments.
