Abstract
Identification of new antigenic peptides, derived from infectious agents, which bind to human leukocyte antigen (HLA) class I molecules, is of importance for the development of new effective vaccines capable of activating the cellular arm of the immune response. In a search for HLA class I restricted antigenic CD8+ T cell (CTL) epitopes, a computer-based program was used to screen and predict the peptide binding affinity for the recently identified twelve HLA class I supertypes. Peptides derived from mycobacterium tuberculosis, vaccinia virus and influenza A protein sequences with a predicted high binding affinity were synthesized, and the binding was biochemically verified and quantified using a recombinant HLA class I molecule-based system.
