Abstract
The aim of the present investigation was to evaluate a non-GMO transformation strategy to breed compact potted plants by using the naturally occurring bacterium Agrobacterium rhizogenes. Recently, it was shown that Kalanchoë blossfeldiana 'Molly' transformed with rol-genes (T1) exhibited a compact phenotype and improved ethylene tolerance. In the present study, the focus was the characterisation of the F1 and F2 progeny. F1 lines were produced by crossings between a selected T1 line and the commercial Kalanchoë cultivar 'Sarah'. F2 populations were produced by self-pollination of selected individual lines. A number of F1 and F2 lines were analysed genotypically. The presence of rol-genes was confirmed in both F1 and F2 plant lines exhibiting dwarfism. In the F1 progenies, the four rol-genes were linked and inherited dominantly in a Mendelian fashion. The segregation of the F2 population is currently being analysed. Phenotypical characterisation in a greenhouse trial revealed that both growth and development were affected. Several F1 and F2 plants exhibited a decreased plant height and changes in flowering and ethylene tolerance. Screening of F2 populations showed a different pattern of distribution of height and ethylene tolerance, depending on the parental lines. Populations derived from WT plants were taller than populations from parents containing rol-genes. The introduction of wild type genes from A. rhizogenes into ornamental plants is a valuable method of molecular breeding to obtain compact growth without using recombinant DNA technology. Compact potted plants can be obtained and lines without delayed flowering and improved ethylene tolerance are valuable for further breeding programmes.
