Abstract
Objective: Interferon-tau (IFNT) secreted by the conceptus is an important factor in the maintenance of luteal function in cows during early pregnancy until day 36. In this multiplex, synepitheliochorial placenta the expression of oxytocin receptor (OXTR) is resumed in the intercaruncular but not in the caruncular endometrium on day 50, although being similar in both regions in cyclic cows. The IFNT protein seems to suppress endometrial estrogen (ER) and OXTR thus inhibiting luteolytic PGF 2 alfa. Our hypothesis was that IFNT synthesis continues in placentomal trophoblast after day 36 regulating the expression of OXTR in the apposed caruncular epithelial cells in a paracrine manner.
Methods: Fifty cows were used for immunohistochemical localization of IFNT and OXTR in bovine placentomes; OXTR mRNA was localized by in situ hybridization. RT-PCR and Western blot were used to confirm local synthesis of IFNT. Sequencing of the PCR product confirmed identity with IFNT1.
Results: The placentomal trophoblast expressed IFNT, OXTR and OXTR mRNA until late pregnancy. The apposed maternal cryptal cells dis not show OXTR protein until late in pregnancy and at parturition, but they expressed OXTR mRNA at all stages examined.Sequencing of the PCR product showed 99 and 97% homology with IFNT1 and Western blot from days 80, 180, and 270 of pregnancy showed a band of 22 dDA, confirming the presence of IFNT.
Conclusion: Bovine plaentomes express IFNT and IFNT1 until near term and thus may regulate OXTR mRNA and/or OXTR expression until parturition. Supported by a grant from the Deutsche Forschungsgemeinschaft to R.I.
